Goncalo Inacio opened the scoring before Pedro Goncalves and a Luis Suarez penalty forced the tie into extra time.
The dramatic comeback was sealed in the additional period as Maximiliano Araujo and Rafael Nel found the net to secure a 5-3 aggregate win.
This landmark victory sees Sporting reach the quarter-finals for the first time since the 1982-83 season.
